Primary open-angle glaucoma (POAG) is one of the leading causes of blindness worldwide. The association between the APOE ε2/ε3/ε4 polymorphism and the risk of POAG has been widely reported, but the results of previous studies remain controversial. To comprehensively evaluate the APOE ɛ2/ɛ3/ε4 polymorphism on the genetic risk for POAG, we performed a systematic review and meta-analysis of previously published studies. The PubMed and Web of Science databases were systematically searched to identify relevant studies. Data were extracted from these studies and odds ratios with corresponding 95% confidence intervals were computed to estimate the strength of the association. Stratified analyses according to ethnicity and sensitivity analyses were also conducted for further confirmation. A total of nine studies were eligible for the meta-analysis, and these studies included data on 1928 POAG cases and 1793 unrelated match controls. The combined results showed that there were no associations between the APOE ε2/ε3/ε4 polymorphism and POAG risk in any of the 10 comparison models. The analysis that was stratified by ethnicity subgroups also failed to reveal a significant association. The sensitivity analysis confirmed the stability and reliability of the findings. There was no risk of publication bias. Our meta-analysis provides strong evidence that the APOE ε2/ε3/ε4 polymorphism is not associated with POAG susceptibility in any populations.  相似文献

Nucleotide sugar transporters play critical roles in glycosylation of proteins, lipids and proteoglycans, which are essential for organogenesis, development, mammalian cellular immunity and pathogenicity of human pathogenic agents. Functional deficiencies of these transporters result in global defects of glycoconjugates, which in turn lead to a diversity of biochemical, physiological and pathological phenotypes. The photosynthetic parameters in leaves of three-year-old seedlings of Fraxinus rhynchophylla L. were studied under different soil water conditions and CO2 concentrations ([CO2]) with a LI-COR 6400 portable photosynthesis system. The objective was to investigate the response of photosynthesis and stomatal conductance (g s) to various [CO2] and soil water conditions, and to understand the adaptability of F. rhynchophylla to such conditions. The results showed that the soil water content (RWC) required to maintain high photosynthetic productivity in F. rhynchophylla was 49.5–84.3%; in this range, net photosynthetic rate (P N) rose with [CO2] increasing from 500 to 1,400 μmol mol?1. Outside this RWC range, P N decreased significantly. The apparent maximum photosynthetic rate (P max,c) and carboxylation velocity (V c) increased with increasing RWC and remained relatively high, when RWC was between 49.5 and 96.2%. CO2 compensation points and photorespiration rate exhibited a trend opposite to that of P max,c and V c, indicating that moderate water stress was beneficial for increasing plant assimilation, decreasing photorespiration, and increasing production of photosynthates. g s declined significantly with increasing [CO2] under different water supplies, but the RWC range maintaining high g s increased. g s reached its maximum, when RWC was approximately 73% and then decreased with declining RWC. The maximal g s was found with increasing RWC. Thus, based on photosynthetic characteristics in artificial, vegetation construction in semiarid loess hill and gully area, F. rhynchophylla could be planted in habitats of low soil water content.  相似文献

DIAPH1 is a formin protein which promotes actin polymerization, stabilizes microtubules and consequently is involved in cytoskeleton dynamics, cell migration and differentiation. In contrast to the relatively well-understood signaling cascades that regulate DIAPH1 activity, its spatial regulation of biogenesis is not understood. A recent report showed that synthesis of DIAPH1 is confined in the perinuclear ER compartment through translation-dependent mRNA targeting. However, the underlying mechanism of DIAPH1 local synthesis is yet to be elucidated. Here, we provide evidence to demonstrate that the 5′-cap-mediated immediate translation of DIAPH1 mRNA upon exiting nucleus is required for localizing the mRNA in the perinuclear ER compartment. This is supported by data: 1) Delayed translation of DIAPH1 mRNA resulted in loss of perinuclear localization of the mRNA; 2) Once delocalized, DIAPH1 mRNA could not be retargeted to the perinuclear region; and 3) The translation of DIAPH1 mRNA is 5′-cap dependent. These results provide new insights into the novel mechanism of DIAPH1 local synthesis. In addition, these findings have led to the development of new approaches for manipulating DIAPH1 mRNA localization and local protein synthesis in cells for functional studies. Furthermore, a correlation of DIAPH1 mRNA and DIAPH1 protein localization has been demonstrated using a new method to quantify the intracellular distribution of protein.  相似文献
